A Seminar Sensitive and Accurate Viral Integration Detection in HBV-associated Hepatocellular Carcinoma by Miss Lyu Xueying Synopsis: Hepatitis B virus (HBV) infection is one of the key etiological risk factors for HCC. Sensitive and accurate detection of HBV integration is crucial to the investigation of the molecular pathogenesis of HCC. We developed the software AVID for HBV integration detection with the enhanced algorithm. AVID outperformed other existing tools in terms of precision, F1 score, and recall in simulated datasets and experimentally validated datasets. Notably, regarding the landscape of HBV integration in HCC, we were able to sensitively and accurately identify recurrent HBV integration events related to TERT, KMT2B, and CCNE1. and Anti-Stanniocalcin 1 Antibody as a Potential Therapeutic Strategy for Liver Fibrosis and Hepatocellular Carcinoma by Mr Suen Long Hin Synopsis: Liver fibrosis is part of the multi-step hepatocarcinogenesis process and persists after the development of hepatocellular carcinoma (HCC). Stanniocalcin 1 (STC1) is a serum biomarker in liver fibrosis and HCC, which is functionally involved in both diseases. In this study, the preclinical efficacy of anti-STC1 antibody in mitigating both metastatic potential of HCC cells and activation of hepatic stellate cells (HSCs) was evaluated. The functional role of anti-STC1 antibody in the interplay between HSCs and HCC cells was also studied. The downstream molecular mechanisms mediated by anti-STC1 antibody in the liver fibrosis cell line model was elucidated by proteomic profiling. All are welcome |

Room 035, 6/F, Block T, Queen Mary Hospital Precision mouse model guides precision medicine in hepatocellular carcinoma - the genetic landscape determines the immune landscape and response to immune checkpoint inhibitorsby Mr. Yuen Wai Hin Vincent, PhD student (Supervisor: Dr. Carmen Wong) Synopsis: "Hepatocellular carcinoma is highly lethal due to limited treatment options. To date, tyrosine kinase inhibitors (TKIs) and immune checkpoint inhibitors (ICIs) are the only two classes of FDA-approved drugs for advanced HCC patients. TKIs have long clinical history in HCC treatment with modest survival benefits. ICIs have remarkable outcome but only limited to a minority of HCC patients. Understanding the mechanisms that determine the efficacy of ICIs in HCC is beneficial to the stratification of patients who will likely respond to ICIs. The primary focus of this study is to elucidate how genetic composition orchestrates the immune composition of HCC. To robustly study how genetic alterations shape HCC tumor immune microenvironment (TIME) thereby affecting the response to ICI, we generated a library of mouse HCCs with genotypes that closely simulate the genetic composition found in human HCCs using genome-editing systems involving transposon and CRISPR-Cas9 systems by hydrodynamic tail-vein injection (HDTVi). Based on the CD8+ T cell infiltration level, we characterized different HCC genotypes into cold tumors (low number of CD8+ T cell) and hot tumors (high number of CD8+ T cell). Anti-PD-1 treatment had no effect in cold tumors but was greatly effective in hot tumors. RNA sequencing of HCC tissues and single-cell RNA sequencing analysis of tumor-infiltrating T cells identified the responsible proinflammatory cytokines and chemokines for CD8+ T cell infiltration in hot tumors. We further characterized different subsets of exhausted T cells in hot and cold tumors and identified a new exhausted T cell population which might potentially serve as a predictive indicator to anti-PD-1 treatment. Interestingly, we found that the TKI, Sorafenib, sensitized cold tumors to anti-PD-1 treatment. In sum, our study has exemplified the use of genome-editing systems to generate mouse HCCs to precisely simulate the same genetic makeup of human HCC to pave the way for precision medicine." Room 035, 6/F, Block T, Queen Mary Hospital (Face-to-face format) LANCL1-FAM49B axis promotes cancer stemness of hepatocellular carcinoma by reducing NADPH oxidase-mediated oxidative stressby Mr. Hongyang Huang, PhD student (Supervisor: Professor Irene Ng and Dr. Yu-Man Tsui) Synopsis: "Cancers including hepatocellular carcinoma (HCC) contain a subpopulation of highly tumorigenic tumor-initiating cells (TICs). Targeting TICs is strategic for cancer treatment. Here, we identify a novel TIC marker of HCC, LANCL1, and its upregulation promotes TIC properties including sphere formation, chemoresistance, and tumor initiation. Our mechanistic study indicates that LANCL1-mediated cancer stemness is via suppression of ROS. LANCL1 blocks the interaction of FAM49B with an E3 ligase, TRIM21, thus protecting FAM49B from ubiquitination and degradation. Enrichment of FAM49B inhibits RAC1 activation, which in turn reduces NADPH oxidase (NOX)-mediated ROS production. Targeting the LANCL1-FAM49B axis may be a potential strategy for HCC treatment." FSP1 is one of the major pathways protect cells against ferroptosis, that is recently discovered. Excitingly, we found that HCC cells relied on FSP1 to protect them against ferroptosis and the induction of ferroptosis by FSP1 inhibitor (iFSP1) suppressed HCC growth. More importantly, iFSP1 altered immune landscape of HCC tumor and promoted tumor infiltration of DCs, macrophages and T cells. iFSP1 also worked synergistically with multiple immunotherapies. In sum, ferroptosis induction promotes innate and adaptive anti-tumor immune responses and is a promising therapeutic approach in treating HCC." All are welcome |

by "Ovarian carcinoma (OC) is a malignancy with grave prognosis owing to frequent resistance to apoptosis-inducing platinum-based chemotherapy. Ferroptosis is a recently defined non-apoptotic cell death driven by the accumulation of iron-dependent lipid reactive oxygen species (ROS), resulting in membrane oxidative damage and cell rupture. Lines of evidence suggested that OC may be susceptible to ferroptosis. However, the effects mediated by ferroptosis and the underlying regulatory mechanisms in OC are not fully characterized. Herein, we demonstrated the general susceptibility of OC cells to ferroptosis and the critical role of ferroptosis in enhancing the cytotoxicity of cisplatin in cisplatin-resistant OC. Additionally, we suggested that EPT1 exerted as a potential ferroptosis suppressor gene by modulating autophagy, providing new insights into the molecular mechanisms of ferroptosis in OC." Targeting CHK2 to Reverse the Tamoxifen (Supervisor: Professor KHOO UI Soon) Synopsis: "BQ323636.1 (BQ) a splice variant of NCOR2, is associated with tamoxifen (TAM) resistance. TAM can cause DNA-damage, triggering the ATM-CHK2 DNA-damage response pathway. We hypothesized this might upregulate BQ-expression, causing TAM resistance. Targeting ATM can cause BQ downregulation, but results in cytotoxicity. Hence targeting CHK2, downstream of ATM, might be a better strategy. Using TAM resistant cell-line LCC2, we confirmed that CHK2-knockdown, or treating cells with CHK2 inhibitor, PV1019, could reduce BQ-expression and sensitize LCC2 to tamoxifen, both in vitro and in vivo. These findings suggest that targeting CHK2 could be a strategy for reducing TAM resistance in breast cancer." All are welcome |

The University Pathology Building (UPB) will be decommissioned in 2018 as part of the Queen Mary Hospital (QMH) redevelopment plan. Since its establishment, UPB has played important role in the undergraduate and postgraduate education of pathology in Hong Kong as well as contributing to breakthrough in research and advancement of clinical services. Besides lectures and practical classes for pathology teaching for medical students, postgraduate courses for medical and related professions are also held in UPB. UPB is also the venue for the annual College examinations. In recent years, educational workshop for the public, particularly high school students, are often conducted in UPB as activities to publicize the International Pathology Day. |
